Separation of proteins is achieved in this instrument using a pH
gradient immobilised on a plastic strip (IEF strips) on the basis
of their isoelectric points. |

Proteins separated on IEF strips are separated in a second dimension
by SDS polyacrylamide gel electrophoresis and are stained with mass
spectrometry compatible Coomassie Blue. |

Linked to the PDQuest software is the spotcutting robot which excises
the spots from the gel of interest. |

MassPrep Station (robotic liquid
handling)
Protein spots are chemically modified and ultimately cleaved using
trypsin an enzyme which cleaves proteins at specific amino acid
residues in the polypeptide chain. This generates a mixture of peptides
whose masses can act as a “fingerprint” of the parent
protein. |
