 Analytical Facilities - Proteomics Section

  Techniques: Garry Rucklidge/ Martin Reid

BioRad_IEF

BioRad IEF cells

Separation of proteins is achieved in this instrument using a pH gradient immobilised on a plastic strip (IEF strips) on the basis of their isoelectric points.

Isodalt

Isodalt & BioRad Dodeca cell large format electrophoresis tanks

Proteins separated on IEF strips are separated in a second dimension by SDS polyacrylamide gel electrophoresis and are stained with mass spectrometry compatible Coomassie Blue.

BioRad_Spotcutter

BioRad spotcutter (compatible with PD Quest)

Linked to the PDQuest software is the spotcutting robot which excises the spots from the gel of interest.

MassPrep

MassPrep Station (robotic liquid handling)

Protein spots are chemically modified and ultimately cleaved using trypsin an enzyme which cleaves proteins at specific amino acid residues in the polypeptide chain. This generates a mixture of peptides whose masses can act as a “fingerprint” of the parent protein.
