The section has 4 gas chromatography
mass spectrometers (GC/MS). These instruments are mainly used to
quantitate compounds of biological interest by isotope dilution
and to measure the enrichment in deuterium, carbon-13 or nitrogen-15
in target metabolites in relation to kinetic studies using stable
isotopes as tracers. These metabolites are isolated from biological
fluids using chromatography techniques prior to derivatisation and
GC/MS analysis. The instruments have the ability to operate in two
modes of ionisation. Chemical ionisation (CI) and electron impact
ionisation (EI). CI is a soft ionisation technique which is used
to enhance the molecular ion, this is a very sensitive technique
requiring only small quantities (pg) of material. Electron impact
ionisation (EI) requires more material (ng) but looks at unique
fragments derived from each metabolite. Using EI allows the isotopic
labelling in different atoms of the molecule to be defined (isotopomer
analysis) and thus can support more complex metabolic studies utilising
multiple tracers. |
