Electrospray ionisation (ESI) is a relatively new technique, which is now the most common method of ionisation used in the analysis of large bio-molecules by analytical mass spectrometry. ESI is a method by which ions, present in solution, can be transferred to the gas phase by applying a voltage, at atmospheric pressure, to the sample droplet. ESI is a soft ionisation technique that permits the analysis of intact molecules at high accuracy (1 part in 10000), and with the propensity to produce multiply charges ions, has an upper mass range of over 150kDa depending on the analysers used. It is one of the most effective interfaces for liquid chromatography (LC/MS) and capillary zone electrophoresis (CZE/MS).

The Institute has three LC/ESI/MS instruments: An Applied Biosystems Q-Trap 2000 triple quadrupole-linear ion trap mass spectrometer with a nano-LC system which supports the Proteomic Section and is used to characterise proteins, an Applied Biosystems API 3200 triple quadrupole mass spectrometer with an Agilent LC system which is used in the quantification of small, organic molecules and an Applied Biosystems Q-Trap 4000 triple-quadrupole-linear ion trap with an Agilent LC system which is also used to quantify small molecules but also has the capabilities to characterise proteins when coupled to a nano-flow system.

ABI_Voyager

ABI Voyager DE PRO MALDI TOF mass spectrometer

The MALDI TOF mass spectrometer measures the masses of the tryptic peptides generated by the MassPrep station. Matching these peptide masses against a database provides protein matches which using bioinformatic packages can provide information about the protein of interest in a particular biological process. (see Proteomics section).
